A-1 Medium can be used in a single step procedure for the detection of fecal coliforms in non-potable water, wastewater and food. Prior enrichment in a presumptive medium is not required. A-1 Medium was formulated to speed up the recovery of E. coli and reduce the incidence of false positive cultures. Gas production in the inverted vial (Durham tube) or dissolved gas that forms fine bubbles when slightly agitated is a positive reaction indicating the presence of fecal coliforms. The medium is designed to favor thermotolerant coliforms, particularly E. coli, by using elevated incubation temperatures (typically 44.5-45.5°C), which suppresses non-target organisms.
